Members of the Order of Ascension are killed for their ascension keystone drops. Gladii are slightly harder than rorarii, and are more spread out, but this is compensated for their high drop rates of noted and alchable items. With high-end gear, you can kill 250 gladii per hour.

When attacking gladii, they will try to move backwards until they reach approximately 6 squares away from you and then charge at you. This attack does no damage, but it will cause a 3 second stun and bind (unless you are still fighting nearby gladii, which will cause a 1 second stun and 3 second bind). To counter this, you can use Freedom or Anticipate to bypass their stuns.

Avoid using Rapid Fire unless Freedom or Anticipation is used against them, as it will cause them to move back rapidly before stunning you, wasting the ability's effect. Fragmentation Shot is a helpful ability when killing gladius, as their constant movement while taking heavy damage will cause the 2x damage effect to activate.

Gladii are usually near capsarii, who are the healers. There are three spots without capsarii, but the gladii there are slightly spread out and can stun you. If one does not mind killing the capsarii, there are a large number of spots to kill gladii. Optional areas for uninterrupted combat are the narrow passageway near the level 70 Agility, the jail section at the bottom left corner or the caverns near Legio Primus' laboratory. For a clearer overview of gladii locations you can use this map.

The gladius's main drops are the ascension keystones, which are believed to be dropped at a 1/64 rate. This means if you kill 250 gladii per hour, you should get around four keystones per hour. They drop a large number of items, especially mid-high level alchables often and mid-high level noted items to compensate for their increased difficulty and numbers.

A spring cleaner or pack yak can be used instead of bringing runes for High Level Alchemy.
